Chris Redfield returns as the main character in Resident Evil 5, a game set five years after Resident Evil 4. He is sent to Kijuju, Africa, where local agent Sheva Alomar joins him on a mission to capture Ricardo Irwing, who is trying to sell bioweapons on the black market. They are soon attacked by hordes of locals infected with an improved version of the "Las Plagas" parasite from Resident Evil 4, which they call "Majini". As the story progresses, Chris discovers that some old acquaintances are behind the whole incident.

This fifth instalment in the series not only carries over the "Las Plagas" parasite concept, but also builds on the innovative gameplay of the previous game. With an over-the-shoulder view, Chris confronts hordes of infected enemies who can throw objects and even shoot at you. They can be fended off with various weapons, grenades, or knife and melee attacks if they get too close. This time, however, Chris has a partner, Sheva, who accompanies him throughout the game and can be controlled by a second player, with both characters needing to watch out for each other in order to survive. The second player can drop in and out at any time, either locally via split screen or online via another console.

Mercenaries mode, consisting of enclosed levels in which the player must survive for a set time while defeating enemies to build up combos and rack up points, also returns from Resident Evil 4.
