The team the player controlled in Tom Clancy's Rainbow Six: Vegas was not the only Rainbow Six unit in Sin City at the time. Under the command of team leader Bishop, a second unit was clearing out the city's back alleys while the other team was enjoying itself in the casinos. The player takes control of Bishop and visits the other, seedier side of Las Vegas.

The gameplay hasn't changed since the first title, so the game again gives the player four teammates he can command through the tactical interface with orders like "Move to" or "Use Gadget". As he moves through enemy-infested areas, gets covered by his team, defuses bombs and rescues hostages, two of the new features become apparent: Persistent Elite Creation (P.E.C.) and Advanced Combat Enhancement Specialisation (ACES).

P.E.C. allows the player to build a unique identity and save their progress from one game to the next. Both in single-player and multiplayer, the player earns experience for killing villains and successfully achieving objectives. Once he has gathered enough experience for the next rank, he unlocks new weapons, body armour and other more or less useful items. The ACES system goes a step further and rewards the player with a large chunk of experience and powerful weapons when he masters challenges in one of three categories: Marksman, Close Quarters and Assault. These challenges range from simply completing a specific story-mode map to a string of consecutive headshots.
